The Khan Lab investigates mechanisms linking inflammation, autophagy, and carcinogenesis in patients with colorectal cancer, focusing on colitis and environmental exposures such as burn-pit toxins. Using murine models, organoids, and advanced molecular techniques, we aim to define how innate immune sensors like cGAS regulate epithelial and immune responses during inflammation-associated colorectal cancer development.
We are seeking a Laboratory Research Technician IV to join our translational research team. The ideal candidate will be experienced in molecular biology, histology, and cell-based assays, capable of managing complex workflows independently, and comfortable in both mouse and human tissue research settings.
Key Responsibilities
Perform and oversee molecular and cellular experiments, including DNA/RNA extraction, PCR, Western blotting, and ELISA
Conduct murine studies including genotyping, sample collection (colon, lymphoid organs), tissue processing, and histology preparation (FFPE, frozen)
Establish and maintain primary epithelial and immune cell cultures, including murine and human colonic organoids, and perform co-culture and drug treatment assays
Execute and assist with immunofluorescence and immunohistochemistry
Assist in flow cytometry and single-cell RNA sequencing sample preparation
Maintain detailed electronic lab notebooks and organize experimental data for analysis and publication
Support the PI and trainees in data analysis, figure preparation, and manuscript writing
Manage laboratory inventory, ordering, and compliance with safety protocols; serve as lab safety liaison as needed
Train and mentor junior staff, students, or fellows in lab techniques
Preferred Skills and Experience
Bachelor's or Master's degree in biological sciences with at least 3 years of laboratory experience, preferably in academic research
Proficiency in cell culture and organoid systems, including sterile technique, cryopreservation, and passaging
Experience with murine handling, tissue harvesting, sample preparation for histology and molecular assays
Hands-on experience with microscopy, immunofluorescence, and image analysis software (ImageJ, QuPath, or similar)
Experience with omics workflows (scRNA-seq, spatial transcriptomics, or related bioinformatics) is an advantage
Excellent organization, recordkeeping, and communication skills; ability to work both independently and collaboratively
